What Is 5-Amino-1MQ and How Does It Work

5-Amino-1MQ is a synthetic small-molecule inhibitor of nicotinamide N-methyltransferase (NNMT), an enzyme expressed at elevated levels in white adipose tissue. Worth noting up front: 5-Amino-1MQ is not a peptide - it's a methylquinolinium small molecule - though it is commonly marketed and sold alongside peptides in the research chemical space, which is why it shows up in peptide-focused catalogs and guides.

NNMT normally diverts nicotinamide away from the NAD+ salvage pathway, converting it to 1-methylnicotinamide (1-MNA) via a reaction that consumes S-adenosylmethionine (SAM). By competitively inhibiting NNMT, 5-Amino-1MQ is studied for its ability to keep more nicotinamide available for recycling into NAD+ through NAMPT, while preserving intracellular SAM. In adipocyte and mouse models, this shift has been associated with suppressed lipogenesis, reduced adipocyte size, and lower body weight - a proposed mechanism for reducing fat cell growth and supporting metabolic rate that does not rely on stimulant activity.
